Take a look at the Event Viewer for the underlying error. It's likely a permission issue.
Please ask your Hosting Provider or your Server Administrator to RESET both Read & Write Permissions for DNN.

Check for license.lic files with file size zero bytes.
Located in DestopModules\LiveModuleName (i.e. LiveTabs).
In IIS Manager, Stop Application Pool. Stop Website. Rename license.lic file to license.lic.old
Restart application Pool, Website. Browse website (might need CTRL + F5)
Should clear error. Verify that license.lic has been recreated!
For skin error on a Mandeeps skin, like Tucson, you may have to restore license.lic from backup (located in portals\_default\skins\Tucson
